Serial dilution of TTX (tetrodotoxin)?
I have a small vial of tetrodotoxin powder, to which I have added 1ml (or 1000 microlitres more precisely) of distilled water. I need to use this as a standard on a HPLC machine, which I have never used before, and to make a calibration curve of its toxicity. How can I serially dilute the 1ml of 0.1mg/ml TTX in a sensible number of 'standards' in order to produce the curve that I need? Also, what would the concentration of each of these dilutions be?
Thanks!
Suggest double - dilute. This would give you a broad range of dilutions.
Neat, 1:2, 1:4.....................1:2048 and beyond if necessary.
Also, you would need to find the linear range by arranging a run-through. Then you will know what dilutions you need to prepare. You will have your curve. Then dilute out your test / QC to bring them into range.
You may find that e.g. 1:8 >>>1:128 is linear and that the test / QC works best at 1:5.
Neat TTX is 0.1 mg/ml,
1:2 TTX is 0.05 mg/ml, (i.e. half),
1:4 TTX is 0.025 mg/ml, (i.e. quarter).
etc.
