Where do lab protocols in molecular biology come from?
Who perfected them? Who invented them? For instance I would like to know the protocol for extracting & purifying Plasmid DNA come from?

You'd probably have to go back to original research articles to find out who was the first to do something and how they did it. Some notable techniques may be written about in books on scientific history.
In general, after a technique is invented, other scientists adapt it to their own needs, making small or drastic revisions. Once something becomes fairly common, you can get protocols from books or they are provided in product manuals (e.g. because Qiagen wants to sell you plasmid purification kits). Often generic protocols can still be improved for a particular circumstance by changing parameters such as concentration, time, or temperature. For routine plasmid prep this isn't necessary, but ELISA assays for instance almost always need a customized protocol.
